Research Focus:
My laboratory uses embryonic stem cells to study the effects of environmental toxicants on prenatal stages of development. Each year over 1,000 new chemicals are released into the environment, most without toxicological testing. Embryonic stem cells are potentially valuable models that can be used to identify chemicals that are harmful to prenatal development and thereby provide information that could be used to prevent disease and congenital defects. While much of our prior work has been done using mouse embryonic stem cells, we are currently adapting our assays to human embryonic stem cells which will provide a unique opportunity to directly examine chemicals for toxicity using human cells.
We have recently completed a study comparing the toxicity of mainstream (MS) and sidestream (SS) smoke from traditional and harm reduction cigarettes on attachment of mouse embryonic stem cells to an extracellular substrate and on proliferation of attached cells. Our results showed that filtering MS smoke significantly reduced its toxicity and that SS smoke is far more potent than filtered MS smoke. Unexpectedly, we found that SS smoke from the harm reduction brands, was more potent in the attachment assay that SS smoke from the traditional brand. This finding alerts us to the potential danger of SS smoke from harm reduction cigarettes and shows that MS smoke from harm reduction cigarettes, while perhaps reduced in certain carcinogens, still retains significant toxicity in assays that measure two fundamental processes in embryos, attachment and proliferation.
We are currently following up these studies with additional assays, testing specific chemicals in smoke, and extending our assays to human embryonic stem cells.
